Every threaded fastener associated with doing a brake job will have a torque spec and possibly call out thread locker. But the torqueing I was primarily referring to is when tightening the lug nuts. If you do not follow the recommended torque pattern and value when installing the wheel, you can end up with a pulsation during braking. Also, you want to be real sure the hub and rotor mating surfaces are free of any debris before installing the rotor. This is less likely to be an issue if using new rotors, but can be quite a concern with old rotors that have flaky rust in the rotor vents. That flaky rust can sometimes come free and end up between the hub and rotor. That typically will yield a pulsation under even light braking at low speeds though.
The specifics on the torque sequence and value can be found in your owner's manual. If you're curious what I do, I snug up all lug nuts by hand first, then turn my cordless impact to the middle setting and go around in a star pattern, only letting the impact hammer for a half a second or so. Then I remove the jackstands and lower the car to the ground, followed by using my torque wrench and torqueing all lug nuts, again using the star pattern, to the recommended value of 100 lbf-ft. Following the proper torque pattern and not significantly over-torqueing is probably most important.
Based on your original description, I really don't think the issue you are experiencing is due to a caliper or hose, unless like I mentioned previously, the vibration/pulsation continues for even a short time after you release pressure from the brake pedal.
