I plan on drag racing my SHO this summer. Am I damaging anything on my car smoking my tires on the line?

I would imagine pushing in the e-brake would reduce the stress on the tranny compared to dropping the clutch to burn out.

I made about 45 passes on the strip with my stock differential, about 10 of those were with drag radials. My differential grenaded and destroyed the transmission case. If you want to run the drag radials just be prepared to possibly break an axle or your differential eventually.

Get a Quife and do all the burn outs you want. The stock dif's wern't made for that kind of use/won't hold up for long.

Yes, the engines were made to handle being raced (not abused though), however the differential will blow up. All I can offer you in addition to everyone else is don't do any sort of excessive wheelspinning. That's what hurts it, if one wheel gets spinning faster than the other for too long (as in a burnout).

I've also heard that the tranny fluid you use affects the durability of the differential. This may be looking into if true.
